摘要
通过研究激素的浓度配比、光照强度以及培养器皿的通透性对叶盘再生的影响,建立了红丰草莓叶盘的高效再生体系。结果表明:叶盘的最适再生培养基为MS+B5(MS无机盐类和B5有机成分)基本培养基附加激素BA 4.0 mg/L、IAA 0.2 mg/L,再生率可达82.5%,单叶盘再生芽数达4.8。另外,预先进行暗处理、控制(培养容器)培养基的通透性均能显著提高叶盘的再生率。
An efficient and reproducible method of direct shoot regeneration from leaf disks in strawberry ( Fragaria x ananassa cv. Hong-feng) was developed. The influence of hormone concentration, light intensity, and ventilation of the container on shoot regeneration was ex- amined. Result showed that regeneration rate of leaf disks could reach 82.5% in MS salts and B5 vitamins medium supplemented with BA 4.0 mg/L and IAA 0.2 mg/L in 60 d, with an average of 4.8 shoots per leaf disk. Dark treatment and fine ventilation of the container improve the regeneration frequency.
