摘要
该文采用聚丙烯酰胺凝胶电泳技术检测了花丝长度不同的三个刺五加类群的异柠檬酸脱氢酶(IDH)、苹果酸脱氢酶(MDH)、过氧化物酶(POD)、细胞色素氧化酶(CCO)和酸性磷酸酶(ACP)五种同工酶型。结果显示,在多态位点0.99标准下,刺五加多态位点比例(P)为0.37.平均每个位点的预期杂合度(He)为0.3200~0.3877,平均每个位点的实际杂合度(Ho)为0.2703~0.3599,刺五加类群内基因多样度(Hs)为0.3387,类群间基因多样度(Dst)为0.0356,类群间基因分化系数(Gst)为0.0951,刺五加三个类群的遗传相似性(I)为0.909~0.941,遗传距离(D)为0.059~0.091。初步推测刺五加是由雌雄同株的两性花逐步向雌雄异株的单性花分化.L-刺五加将进化成单雄性.S-刺五加将进化成单雌性。其遗传分化的时间约为29.5~45.5万年。
The phenotypes of five enzymes, Isocitric dehydrogenase,Malate dehydrogenase,Peroxidase,Cytochrome oxidase and Acid phosphatase in three Eleutherococcus senticosus populations with different filament length were analysed by polyacrylamide gel electrophoresis. The results indicated that under 0.99 standard of proportion of loci polymorphic (P), the P of Eleutherococcus senticosus is 0. 31. Its mean expected heterozygosity per locus (He) is 0. 320 0~0. 387 7. Its mean observed heterozygosity per locus (Ho) is 0. 270 3 ~0. 359 9. The gene diversity in every population of Eleutherococcus senticosus is 0. 338 7.The gene diversity among populations of one is 0.035 6. The coefficient of gene differentiation among populations of one is 0. 095 1. The gene similarity coefficient of three populations of one is 0. 909~0. 941 and the gene distance is 0. 059~0. 091. That the Eleutherococcus senticosus is gradually differentiating from hermaphrodite to dioecism is preliminarily inferred. and the L-one will be changed into androecious. the S-one into monogynous. The time of gene differentiation of one is about 295 000~455 000 years.
