摘要
为了便于对生产上主要推广的4个石榴(Punica granatum L.)基因型进行苗木纯度检测,维护育种者和果农的利益,用225对SRAP(Sequence Related Amplified Polymorphism)引物组合对其幼嫩叶片DNA进行了分析。结果发现其中19对SRAP引物组合的23个差异迁移位点的扩增条带稳定、清晰,可以将4个石榴基因型完全区分开,作为其鉴别的标记。将这些条带进行数据化记录和分析,发现仅用ME8/EM11和ME9/EM18位点的共计3个差异迁移位点的数据即可将供试的4个材料区分开。将这些数据输入Excel数据表格,获得了由系统自动生成的4个石榴基因型"分子身份证"号码,可作为这4个石榴基因型间的鉴定依据。
For the convenience of checking on the nursery stock purity of four pomegranate (Punica granatum L.) genotypes which were popularized in production and for the protection of the benefit of breeders and growers, two hundred and twenty-five SRAP(Sequence Related Amplified Polymorphism) primer pairs were used to analyze the DNA from the tender leaves of these four pomegranate genotypes. Amplification products were analysed by electrophoresis on 8% Non-denaturing polyacry-lamide gel and then checked by silver staining. The results suggested that twenty-three discrepant alleles amplified by nineteen SRAP primers were stable and clear, which could be used to identify the four pomegranate genotypes. The results also showed that only using three alleles amplified by ME8/EM11 and ME9/EM18 could differentiate this four materials. The data from the three alleles were entered into an Excel program and then the system automatically create the molecular identity card numbers of four pomegranate genotypes which could be used to identify the four genotypes.
