摘要
玉米淀粉修饰基因du是玉米品质改良的重要资源之一。本研究运用RAPD技术,以4种不同遗传背景的近等基因系(A632 Du/Du、A632 du/du;Mol7 Du/Du、Mol7 du/du;Oh43 Du/Du、Oh43du/du;W64Du/Du、W64du/du)为筛选材料,采用(7922Du/Du×Mol7du/du)和(HZ32Du/Du×Mol7du/du)F_2群体,研究du基因的分子标记。两个群体分别是87和69个单株。共筛选356个随机引物,发现了3个RAPD标记与du基因紧密连锁。运用Joinmap软件的统计分析表明OPA16~0.4kb、OPKll~1.7kb在(7922Du/Du×Mol7du/du)F_2群体与Du基因紧密连锁,遗传距离分别为6.97cM和4.33cM。OPX 9~0.35kb在(HZ32Du/Du×Mol7du/du)F_2群体与Du基因紧密连锁,遗传距离为4.17cM。
Starch-modified gene du is one of important genetic resources in starch genetic modification for maize. The objective of the research was to find molecular markers linking with du gene by RAPD. A total of 356 random primers were screened with 4 iso-genic lines(A632 Du/Du, A632 du/du; Mol7 Du/Du, Mol7 du/du; Oh43 Du/Du, Oh43 du/du, W64 Du/Du, W64 du/ du). Two mapping populations, (7922 Du/Du×Mo17 du/du)F2 and (HZ32 Du/Du×MO17 du/du)F2, were used, among which, 87 and 69 individuals were included respectively. 4 primers produced RAPDs between iso-genic lines. Three of them, OPA16-0. 4 kb, OPK11-1. 7 kb and OPX9-0. 35 kb, were tightly linked with du gene according to statistical analysis. Based on joinmap analysis, map distances of du gene to OPA16-0. 4 kb and OPK11-1. 7 kb were 6. 97 cM and 4. 33 cM respectively. There was 4. 17 cM of map distance between du and OPX-0. 35 kb.
