摘要
目的:探讨不同pH值缓冲液配制出的姬氏染液对血涂片染色效果的影响。方法:使用pH值分别为6.4、6.6.6.8、7.0、7.2、7.4、7.6的缓冲溶液配制的姬氏染液和双蒸水配制的姬氏染液对小鼠血涂片进行染色,比较染色效果。结果:pH值为6.4和6.6时。红细胞清晰着色好,白细胞细胞棱着色淡;pH值为6.8的染色后,红细胞清晰着色好,白细胞可见紫色细胞值;pH值为7.0的染色后,红、白细胞着色均较淡;pH值为7.2的染色后,红细胞清晰着色好,白细胞核清晰;pH为7.4的染色后,红、白细胞均较清晰且着色好;pH为7.6的染色后,红细胞颜色偏蓝,白细胞着色好;用双蒸水配制的染液染色后,红、白细胞均清晰且着色好。结论:低pH值的染液红细胞很好的着色,但白细胞的胞核着色偏淡。而高pH值的染液可使白细胞染色效果较好,但是红细胞着色不佳;使用双蒸水配制的染液,红细胞和白细胞的染色效果都很好。
[ Objective] To study the influence on the staining of blood smear by Giemsa stain of buffer solution with various power of hydrogen. Method : Tested 7 kind of Gimsa made by phosphate buffer in PH6.4, 6.6, 6.8, 7.0, 7.2, 7.4 and 7.6. And the Gimsa made by distilled water as well. Then we observed. Result: We found that the red - blood - cell can be dyeing well by the Giemsa in PH6.4 and PH6.6, and the nucleus of leucocyte are purple; Dyeing by the Giemsa in PH7.0, both erythrocyte and leucocyte are chmmatosis indifferent. When we adjust the PH of Giemsa into 7.2, erythrocyte and nucleus of leucocyte are clearly. And dyeing well by the Giemsa in PHT. 4 either red - blood - cell or white - blood - cell. Staining by Giemsa in PHT. 6, the leucocyte chtomatesis well but the color of the erythrocyte is a little blue. The Gimsa made by distilled water is the best one in this experiment, either for etythrocyte or leucocyte. Discussion : Red - blood - cell can be dyeing well by Giemsa in lower PH, but so indifferent of the color of the leucocyte's nucleus. On the other hand ,staining by the Giemsa in higher PH, we get clear leucocyte and poor staining of erythrcyte. Giemsa made by distilled wate could dyeing both erythrocyte and leucocyte very weLl.
