摘要
目的:探讨石斛对胃热证大鼠的作用及其机制。方法:用干姜水煎剂连续灌胃大鼠15 d,复制胃热证大鼠模型,再以石斛水煎剂(1.5,0.75 g.kg-1)灌胃治疗10 d。比较大鼠的饮食量、大小便、舌色舌苔变化,用红外热扫描成像(therm altexture m aps,TTM)系统检测大鼠全身不同部位的温度改变,用放射免疫分析法测定大鼠血液中与胃机能相关的生化指标血栓素B2(TXB2)、6-酮-前列腺素F1α(6-keto-PGF1α)、胃动素(MTL)、胃泌素(Gas)、生长抑素(SS)、白细胞介素-4(IL-4)、白细胞介素-8(IL-8),观察大鼠胃黏膜组织病理学变化情况。结果:造模后大鼠呈现舌红苔黄(P<0.05)、饮食量增加(10%以上)、大小便量减少(10%以上)、大便多数干结、小便颜色加深(P<0.01),大鼠的头、颈、左腋下、胸、上腹、中腹部体温显著升高(差值>0.5℃或差值>1.0℃);大鼠血液中显著降低的有6-keto-PGF1α(P<0.01),显著升高的有MTL,Gas,IL-8(P<0.01);大鼠胃黏膜受到损伤,呈现炎性变化(P<0.05,P<0.01)。石斛治疗后除食量,头、颈、胸部温度,血浆MTL浓度,胃黏膜腺体主细胞数外,上述胃热证的其他指标均得到改善(差值>0.5℃或差值>1.0℃,P<0.05,P<0.01)。结论:石斛通过使胃热证大鼠血液中的IL-8,Gas,TXB2降低,6-keto-PGF1α升高,改善胃热证大鼠的全身体征,发挥对大鼠胃黏膜的保护作用。
Objective: To observe the effects of Herba dendrobii on rats with stomach-heat syndrome and to explore the mechanisms. Method: Rats were fed with decoction of Rhizoma Zingiberis for 15 continuous days to induce the model of stomach-heat syn- drome. After modeling, Herba Dendrobii (HD) decoction were given (in the doses of 1.5, 0. 75 g · kg^-1 respectively) for 10 days. After treatment, amount of the daily diet, volume and absorbance of urine, pellet number and moistness of excrement, color and coat ing degree of tongue were recorded; the body thermal effects were detected with thermal texture maps (TrM) system; the biochemical indexes of blood reflecting the physiological function of stomach, including thromboxaneB2 (TXB2), 6-keto-prostaglandin F1α (6-keto- PGF1α ), motilin ( MTL), gastrin ( Gas), somatostation ( SS), interleukin-4 ( IL-4 ) and interleukin-8 ( IL-8 ) were measured by radioimmunoassay; and the histological changes of gastric mucosa were observed by hematoxylin-eosin (HE) stain. Result: The model rats had yellow coating and red tongues (P 〈0. 05). The amount of daily diet were increased ( over 10% ), urine volume and excrement pellet number were decreased (over 10% ). The their urine color became deep (P 〈 0. 01 ) and their excrement became dry. The temperatures in head, neck, left fore-armpit, chest, up-abdomen, mid-abdomen of the model rats were raised up ( difference 〉 0. 5 ℃ or difference 〉 1.0 ℃ ). The content of 6-keto-PGF1α in blood of model rats decreased evidently (P 〈0. 01 ), and the contents of MTL, Gas and IL-8 increased conspicuously (P 〈0. 01 ). The histological changes of gastric mucosa in the model rats were as follows: diffuse congestion, infiltration of neutrophil, less secretion, decrease of the number of chief and parietal cells, etc (P 〈 0.05 or P 〈 0. 01 ). After treatment with HD, except the daily food weight, the temperatures in head, neck and chest, the content of MTL and the number of chief cells, the other indexes observed above were improved noticeably ( difference 〉 0. 5 ℃ or difference 〉 1.0 ℃, P 〈0. 05 or P 〈 0. 01 ). Conclusion: The reason why HD relieves the general symptom and sign the gastric mucosa of rats with stomach-heat syndrome is that HD can increase 6-keto-PGF1αand decrease IL-8, Gas, TXB2 in their blood.
