摘要
目的:观察白花前胡甲素对豚鼠心肌细胞电生理特性的影响,以探讨其抗心律失常的作用机制。方法:实验采用标准玻璃微电极技术。结果:PdⅠa对豚鼠心肌细胞的动作电位时程(APD)及慢反应动作电位时程均有缩短作用,PdⅠa还使慢反应动作电位的振幅(APA)明显减小,两者均呈现剂量依赖关系。结论:PdⅠa的抗心律失常作用,可能与阻滞钙离子内流。
OBJECTIVE:To clarify the mechanism of action of Pd Ⅰa,an effective component of Peucedanum praeruptorum Dunn in the treatment of arrhythmia.METHODS:Intracellular microelectrode techniqne was used.RESULTS:With 5 μmol·L -1 and 50 μmol·L -1 Pd Ⅰa,APD 30 was reduced from(122.63±6.16)ms to (113.46± 5.14 )ms and (107.22±6.27)ms(n=8,P< 0.01);APD 50 ,from (145.28±5.71)ms to (138.08±5.92)ms and ( 129.06 ±7.69)ms(n=8,P<0.01);APD 100 ,from (197.39±4.45)ms to (188.23±5.52)ms and (178.26±6.27)ms(n=8,P<0.05)respectively.The inhibitory rates were 12.6%,11.2%,and 9.7%respectively.APD 30 was the most marked one.There were no evident changes on APA and V max .The reduction of APA was also observed in slow action potential(SAP) after adding Pd Ⅰa,and also showed in a dose dependent manner.With(5 μmol·L -1 ) Pd Ⅰa,APA changed from(90.8±3.07)mV to (87.2±3.51)mV,with(50 μmol·L -1 ) Pd Ⅰa to (85.4±3.44)mV(n=5,P<0.01).CONCLUSIONS:The reduction of APD 30 and the inhibition of APA in SAP are considered to be related to the suppression of I Ca .So Pd Ⅰa may have Ca 2+ channel clock effect.
