Dysregulation of polycomb group protein Bmi-1 expression has been linked with an invasive phenotype of certain human cancers and poor prognosis of patients; however, the underlying mechanisms are poorly understood. Here, we demonstrate that Bmi-1 expression is inversely correlated with E-cadherin expression in various cancers. We find that overexpression of Bmi-1 causes epithelial to mesenchymal transition (EMT), enhances cell motility/invasiveness, and contributes to the promotion of tumorigenesis and metastasis; whereas silencing Bmi-1 expression in cancer cells reverses EMT and reduces metastatic potential.
