糖尿病大鼠膀胱超微结构改变及毒蕈碱受体基因表达的研究 被引量:1

摘要 目的:研究病程3周的2型糖尿病(DM)大鼠膀胱超微结构改变以及毒蕈碱型受体基因表达改变。方法:2日龄雌性Wistar大鼠随机分成2型糖尿病组和正常对照组,链脲佐菌素按90mg/kg体重腹腔注射并结合高糖高脂饮食诱导2型糖尿病大鼠模型。于糖尿病病程第4周时观察糖尿病大鼠膀胱超微结构改变;并以RT-PCR法检测毒蕈碱型受体mRNA表达水平。结果:病程第4周时糖尿病大鼠膀胱肌层电镜下有明显的肌细胞核固缩、细胞器空洞样变、细胞连接纤维化、成纤维细胞变性及胶原纤维排列紊乱。糖尿病组大鼠膀胱逼尿肌毒蕈碱型受体mRNA的数量显著高于正常对照组,为(58.0±6.2)%vs(34.8±9.8)%,P<0.05。结论:本研究证实了糖尿病大鼠膀胱超微结构及毒蕈碱型受体表达的改变,为预防、治疗DM所致膀胱病理改变,解决糖尿病性神经源性膀胱提供科学依据。
