摘要
目的:研究微量元素硒和维生素E对三种需谷胱甘肽抗氧化酶活性的保护作用。方法:体外培养仓鼠幼肾成纤维细胞系(BHK—21/C13),在分别加入Fe—ADP、特丁基氢过氧化物(t—BHP)和H_2O_2的情况下,然后同时加入维生素E或微量元素硒,观察三种酶活性的变化。结果:三种促氧化剂均显著刺激MDA生成,抑制谷胱甘肽过氧化物酶的活性,而对谷胱甘肽还原酶无影响。其中t—BHP的作用最强,并可使谷胱甘肽S-转移酶的活性降低。两种抗氧化剂均对上述过氧化反应及酶活性抑制有明显预防作用,且维生素E更为有效。结论:GSHpx较易受到过氧化反应损伤;维生素E是更为有效的抗氧化性保护物质。
Objective: To study the effect of selenium and a - tocopherol on three GSH - requiring antioxidant enzymes. Method: The engyme activities in BHK - 21/C13 were assayed in condition of oxidative stress by Fe - ADP, t-butyl hydroperoxiade (t - BHP) and H2O2, with or without selenium or a-tocopherol supplementation. Results: All three peroxidants stimulated MDA production and inhibited glutathine peroxidase(GSHPx) activity without any affection on glutathine reductase, while glutathine S - transferase activity was decreased by t - BHP only. And t -BHP showed the strongest effect among the three peroxidants. It has been fonud that both selenium and a-toropherol could protect against both peroxidation and enzyme inhibition by peroxidants with more effetive efect offered by α - tocopherol. Conclusion: GSHPx can be damaged by peroxidation and α -tocopherol is one of the powerful antioxidant.
