摘要
为了提高藏獒的繁殖力,建立完善的藏獒人工授精体系和保存优秀藏獒精子,本研究对8只藏獒进行了采精和精液的超低温冷冻保存试验,并对藏獒的精液超低温冷冻保存液进行了系统的筛选。结果表明,渗透压450mOsm/L的配方Ⅴ(葡萄糖1.98g、蔗糖3.42g、柠檬酸钠0.59g、谷氨酸钠0.75g、三羟甲基氨基甲烷1.21g、VB610mL、VB1210mL、卵黄20mL和甘油8mL)对藏獒精液冷冻保存的效果最佳,解冻后精子的活力为0.58,畸形率为17.2%,满足输精和进行超低温冷冻保存精子的要求。
In order to increasing the reproductive capacity of Tibet-mastiff, perfecting artificial insemination(AI)technology and preserving spermatozoa of excellent Tibet-mastiff. We had collect and frozen the semen of eight Tibet-mastiffs, at the same time, had a systemic filtrate to the spermatozoa freezing-extender of Tibet-mastiff. The result showed that the extender Ⅴ (composing by glucose 1.98 g, sucrose 3.42 g, citric acid sodium 0.59 g, glutamic acid sodium 0.75 g, EDTA-Na2 1.21 g, vitamin B6 10 mL, vitamin B12 10 mL,yelk 20 mL and glycerol 8 mL) is the best cryopreservation extender of Tibet-mastiff. The forward motility sperm is 0.58, pathomorphology sperm is 17.2%, meeting the quality criteria apply for AI and storage at -196℃.
