IL-8在牙周膜牵张成骨快速移动牙牙髓中表达变化的实验研究 被引量:2

Immunohistochemical expression of IL-8 in the pulpal tissue on orthodontic rapid tooth movement through distraction osteogenesis of the periodontal ligament

摘要 目的:通过建立牙周膜牵张成骨动物模型,观察牵引后实验动物移动牙牙髓中IL-8表达的变化,并从这一角度探讨牙周膜牵张成骨快速牙移动的生物学机制。方法:本地健康杂种犬10只,随机分为5组,1个对照组,4个实验组。拔除下颌双侧第二前磨牙,并于其远中牙槽中隔进行减阻处理,以单根的第一前磨牙作为移动牙,双根的第三前磨牙作为支抗牙,实验组动物通过牙周膜牵张成骨快速移动牙齿,并分别在加力4W及加力后保持1W、2W、4W时结束实验,局麻下拔除移动牙,取出牙髓组织标本,用ELISA夹心法测定每组移动牙牙髓中IL-8的含量及其在矫治过程中的变化。结果:加力4W后(A组),实验犬移动牙牙髓中IL-8含量与对照组相比显著增高,差异有统计学意义。加力停止1W后(B组),IL-8含量略有下降。加力停止2W后(C组),IL-8含量显著降低至(3.5523±0.3313)pg/mg,与加力4W组(A组)及对照组相比均有明显统计学意义。加力停止4W后(D组)IL-8含量显著下降,与对照组(E组)相比无统计学差异。结论:本实验采用拔牙间隔骨实施微小减阻措施及持续快速牵张的方法,使牙齿移动速度得到较显著的提高,IL-8含量的变化也提示未出现牙髓组织坏死、发生不可逆性炎症等副作用。证实牙周膜可以快速牵张成骨,加快牙齿移动速度,并且无明显不良反应,为临床正畸移动牙齿提供了一种新思路、新方法。 Objective: To research the immunohistochemical expression of IL-8 in the pulpal tissue through establishing the animal model of orthodontic rapid tooth movement through distraction osteogenesis of the periodontal ligament, and discuss the biologic mechanism.Method: Ten native-healthy-hybrid dogs were used.The animals were randomly divided into 5 groups, four groups were according to experimental times and one group was the control group.On the experiment groups, the frist premolar were moved by using distraction device, the third premolar were as anthor teeth.The experiment groups were executed when device was activated 4 weeks (group A), the device was fixed for I week (group B),2 weeks (group C). 4 weeks (group D) after activating respectively. Then immunohistochemical expression of IL-8 in the pulpal tissue was analysized by ELISA method. Result: The IL-8 concentrations in group A were significantly higher than the control group. After the device was fixed for 4 week, the concentrations declined appreciably. The IL-8 concentrations declined markedly, there was significant difference with the group A, no significant difference with the control group.Conclusion: The immunohistochemical expression of IL-8 in the pulpal tissue shows distraction osteogenesis of the periodontal ligament is safe.The distraction through periodontal ligament could induce more rapid tooth movement.
