反复海马CA1区惊厥阈下电刺激致实验大鼠持续性临床下痫样放电研究

摘要 目的:通过海马CA1区惊厥阈下电刺激建立可持续较长时间、无明显惊厥行为的临床下痫样放电动物模型。方法:选用雄性SD大鼠25只,通过反复大鼠海马CA1区惊厥阈下电刺激,观察大鼠惊厥行为,纪录电刺激停止后1、3、7、14、30 d时大鼠皮层脑电图改变。结果:采用刺激频率30 Hz,波宽1 ms,串长10 s,刺激强度0.1 mA,串隔5 min,10次/d,连续刺激4 d,最后一次电刺激后30 d内,均可在大鼠皮层记录到稳定发放的散在或阵发性高幅尖波、棘波、棘慢波,而实验动物并无明显的惊厥行为表现(Racine 0级)。结论:反复海马CA1区惊厥阈下电刺激可较长时间诱发实验动物出现无明显惊厥行为的临床下痫样放电。 Objective: To establish an animal model of persistent subclinical epileptiform discharges (SEDs) induced by repetitive subthreshold electrical stimulation to hippocampus CA1 region without obvious seizure activity in rats. Methods:Electrical stimulation was given to the bippocampus CA1 region in 25 male Sprague Dawley (SD) rats. The seizure activity of experimental rats was evaluated by Racine scores, and electrocorticogram (ECOG) was recorded at 1, 3, 7, 14 and 30 d after the last electrical stimulation. Results:The experimental rats were given subthreshold electrical stimulation to the hippoeampus CA1 region 10 times per day for 4 d through a constant pulsating current with intensity of 0. 1 mA, frequency of 30 Hz, wave width of 1.0 ms, train duration of 10 s,interstimulus intervals of 5 min. Sporadic or paroxysmal epileptiform discharges such as high amplitude sharp or spike waves, or spike and slow wave complex were recorded in 30 days after the last stimulation. Meanwhile, no obvious seizure activities were observed (Racine 0) in experimental rats. Conclusion.. Persistent subclinical epileptiform discharges without obvious seizure activity can be provocated by repetitive subthreshold electrical stimulation to hippocampus CA1 region in rats.
