汽巴蓝纳米微球的制备及与人血清白蛋白相互作用的研究

摘要 利用苯乙烯单封端的聚醋酸乙烯酯(PVAc)大分子单体与苯乙烯进行分散共聚,制得了表面光滑、直径在500~700 nm之间、具有较好单分散性的聚苯乙烯接枝PVAc(PSt-g-PVAc)核壳结构复合微球.在碱性条件下醇解得到PSt-g-PVA微球,进而由活性染料汽巴蓝(CB)与微球表面的羟基进行亲核反应,制得了汽巴蓝功能微球(CB微球).经元素分析测定得到CB微球中CB的含量为139.22μmol/g.该CB微球对人血清白蛋白(HSA)的吸附量为40.9 mg/g.由硫氰酸钠进行解吸试验,吸附在微球上的HSA的解吸率为92.11%,且CB微球的重复使用性能良好. The PSt-g-PVAc microspheres were obtained by dispersion copotymerization of styrene (St) and PVAc macromonomer with a vinylbenzyl end group. And diameters of the PSt-g-PVAc microspheres are 500-700 nm. PSt-g-PVA composite microspheres were derived from alkaline hydrolysis from PSt-g-PVAc microspheres. The functional microspheres were prepared via the nucleophilic reaction between PSt-g-PVA microspheres and Cibacron Blue F3GA (CB). The biggest amount of CB on surfaces of the functional microspheres was 139.22 μmol/g which measured by using an elemental analysis instrument. The amount of HSA adsorption on functional microspheres was 40.9 mg/g. The highest desorption ratio (92.11%) was achieved by using an eluent of 0.5 mol/L NaSCN (pH 8.0). The functional microspheres could be reused without significant decreases in the adsorption capacities.
