摘要
本文研究过氧钒烟酸络合物(POV)对链脲佐菌素(STZ)诱导的糖尿病大鼠肝细胞质烯醇丙酮酸磷酸羧激酶(PEPCK)基因表达及血脂代谢的影响,并探讨POV降低高血糖和高血脂的分子生化机理。结果显示:糖尿病鼠肝细胞质PEPCK基因表达及活性明显增加,总胆固醇和甘油三酯也明显高于正常,用POV治疗后上述变化得到纠正。提示POV的作用机理之一是通过抑制PEPCK基因表达及酶活性减少糖异生而降低高血糖。
The effect of peroxovanadate_nicotinic acid (POV)on gene expression and enzymatical activity of hepatic phosphoenolpyruvate carboxykinase (PEPCK) and blood_lipid metabolism of STZ_induced diabetic rats was investigated and the molecular biochemical mechanism of POV in lowering hyperglycemia and hyperlipemia was also explored. The results showed that the gene expression and enzymatical activity of hepatic PEPCK as well as total cholesterol and triglyceride were significantly increased in diabetic rats with hyperglycemia and hyperlipemia. These changes were corrected with POV treatment. These results suggest that one of the mechanism of POV is to inhibit PEPCK gene expression and PEPCK activity, thereby decreasing glyconeogenesis and hyperglycemia. In the meantime it may promote the recovery of diabetes through modulating lipid metabolism.
