摘要
目的研究CYP2E1基因多态性与中国四川汉族人群肺癌遗传易感性之间的相关性。方法应用PCR-RFLP技术检测150例中国四川汉族肺癌患者和152例健康人的CYP2E1基因RsaⅠ/PstⅠ和DraⅠ多态性的分布频率,并分析了这两种基因多态性与中国四川汉族人群肺癌遗传易感性之间的相关性,以及与吸烟在肺癌易感性中的交互作用。结果①CYP2E1基因RsaⅠ/PstⅠ和DraⅠ多态基因型分布频率在2组间比较无显著性差异(分别为Х^2=3.186,P=0.203和Х^2=1.756,P=0.416);②按照吸烟因素分层,携带c1/c1基因型的不吸烟个体较携带突变基因型的个体肺癌风险显著增加(OR=2.453,95%CI=1.140~5.276,P=0.022);③与携带至少1个突变c2基因型的个体比较,携带c1/c1基因型的个体患肺腺癌的风险显著增加(OR=2.440,95%CI=1.235~4.820,P=0.01);④没有发现DraⅠ多态性与肺癌风险之间的相关性。结论①CYP2E1的c1/c1基因型为中国四川汉族人群肺癌易感基因型;②RsaⅠ/PstⅠ多态性与中国四川汉族人群患肺腺癌的风险显著相关;③DraⅠ多态性与中国四川汉族人群肺癌风险无相关性。
Objective To evaluate the correlation between the polymorphisms of CYP2E1 gene and the susceptibility of lung cancer in Chinese population. Methods Polymorphisms of CYP2E1 RsaⅠ/PstⅠ and DraⅠ were detected in 150 patients with lung cancer and 152 healthy controls in Chinese Sichuan Han subjects by PCR-RFLP. The associations between genetic polymorphisms and susceptibility of lung cancer in Chinese Sichuan Han subjects were analyzed. The interaction among genotype, smoking status and lung cancer susceptibility were also analyzed. Results (1) No significant difference in the frequencies of CYP2E1 RsaⅠ/PstⅠ and DraⅠ genotypes were found between the two groups (Х^2 = 3. 186,P = 0.203 and Х^2 = 1. 756, P = 0.416 respectively) ;(2) Considering the smoking status, more risk of lung cancer was detected in the nonsmokers carried with c 1/c 1 genotype compared with those mutant genotypes ( c 1 and c2 ) carriers ( OR = 2.453,95 % CI = 1. 140 -5. 276, P = 0.022) ; (3) More risk of adenocarcinoma was found in the individuals who carried with c 1/ c1 genotype than in those carried with at least one c2 genotypes ( OR =2. 440, 95% CI = 1. 235 -4. 820, P = 0. 01 ) ; (4) No remarkable association between polymorphisms of CYP2E1 DraⅠ genotypes and the susceptibility of lung cancer were found. Conclusion (1) CYP2E1 cl/cl genotype is susceptible genotype for lung cancer in Chinese Sichuan Han population in China; (2) The polymorphisms of CYP2E1 RsaⅠ/PstⅠ are closely associated with the increased risk for adenocarcinoma in Chinese Sichuan Han population; (3) No association between CYP2E1 DraⅠ polymorphism and lung cancer risk is found.
