中国两性生殖卤虫早期胚胎发育蛋白的双向电泳条件优化

Optimization of conditions of two-dimensional gel electrophoresis of protein from embryo of Artemia sinica

摘要 采用双向电泳技术对中国卤虫胚胎发育到5 h时的蛋白组进行研究与分析.考察不同条件对双向电泳结果的影响,分别从上样量、染色方法、分离胶浓度等3方面进行研究.实验结果显示上样量为400μg时图谱的斑点数较多;采用银染法所得实验结果优于考染;同时分离胶浓度为10%时所得蛋白斑点数较多.优化的实验条件为进一步研究中国卤虫蛋白组奠定基础. Using two-dimensional gel electrophoresis(2-DE), the changes of 2-DE pattern of protein from 5 hour stages of embryo of Artemia sinica were investigated and analyzed. Three different condi- tions, which are loading volume, staining method lyzed. The results show that the spot number was Also,more protein spots were observed from silver and the concentration of separating gel were anaimproved by sample loading volume with 400 μg. staining than Coomassie brilliant blue dying. Moreover,separating gel at lower concentration(10%T) could increase spot volume. The improved experimental conditions lay a better foundation for research in proteomes of Artemia sinica embryo.
