摘要
目的分析胶质瘤细胞分泌基质金属蛋白-2(MMP-2)水平和丝状肌动蛋白(F-actin)的含量,探讨胶质瘤细胞迁移与溶解基质的能力及其与侵袭性的关系。方法分别用流式细胞术、免疫细胞化学法和transwell体外侵袭实验检测胶质瘤细胞和星形胶质细胞F-actin的含量、MMP-2蛋白的表达水平及侵袭力的强弱,并对两种细胞进行比较分析。结果胶质瘤细胞F-actin的含量(202.54±11.06)明显高于星形胶质细胞F-actin的含量(62.64±10.23).两者比较有显著性差异(t=-26.258,P=0.000);胶质瘤细胞MMP-2的表达呈阳性或强阳性.星形胶质细胞MMP-2的表达呈阴性或弱阳性,两组MMP-2阳性表达率分别为96.11%及15.05%.比较有显著性差异(P=0.000);胶质瘤细胞的侵袭力(246.97±54.48)明显强于星形胶质细胞的侵袭力(39.77±11.46).两组比较有显著性差异(t=20.376.P=0.000)。结论胶质瘤细胞含有大量的F-actin及分泌大量的MMP-2蛋白,高F—actin含量与大量MMP-2蛋白分泌与肿瘤细胞强侵袭力有关。
Objective To determine the level of MMP-2 secreted and the content of F-actin in glioma cells and to investigate the abilities of migration and dissolving matrix of glioma cells and the relationship with the invasion. Methods The content of F-actin, the expression of MMP-2 and the invasiveness of glioma cells and astrocytes were detected respectively by flow cytometry, immunocytochemistry and transwell invasion experiment in vitro. Then, the results of the two kinds of cells were compared. Results The content of F-actin in glioma cells (202.54±11.06) was significantly higher than that in astrocytes (62.64± 10.23, t=-26.258, P=0.000). The expression of MMP-2 in glioma cells displayed positive or powerful positive, while it was negative or weakly positive in astrocytes; the positive rate in glioma cells (96.11%) was significantly higher than that in astrocytes (15.05%, P=0.000). Moreover, the invasiveness ofglioma cells (246.97_±54.48) was more powerful than that ofastrocytes (39.77±11.46, t=20.376, P=0.000). Conclusion In contrast to astrocytes, glioma cells possess more F-actin and secrete more MMP-2, which is associated with the invasiveness of glioma.
