股骨头磷酸二酯酶5基因的检测

摘要 【目的】检测股骨头内及其支配血管磷酸二酯酶5(PDE5)基因及其表达水平,为研究磷酸二酯酶5抑制药可选择性增加股骨头血流量提供合理解释。【方法】采用RT-PCR法、Western印迹法和免疫组化染色检测17只雄性狗股骨头、主要髋外周动脉的PDE5基因及其表达。【结果】狗股骨头和髋外周动脉组织提取物用琼脂糖凝胶电泳均可显示较清晰的总RNA条带,分别为28S,18S,5S;RT-PCR扩增PDE5 cDNA后可见大小为426 bp的PDE5特异条带;Western印迹法显示狗股骨头在相对分子质量Mr≈95×103处有PDE5蛋白的表达;免疫组化染色显示狗股骨头PDE5分布于骨髓粒细胞系胞浆内。【结论】股骨头和供血血管有PDE2的正常表达,提示PDE5特异性抑制药可通过调控基因表达而影响股骨头血流量。
