摘要 目的考察Combretastatin A4 phosphate(CA4P)溶液的光解机制及光解动力学规律。方法在不同光源下对CA4P溶液进行加速实验,HPLC测定含量。结果不同浓度溶液在相同光源下的光解动力学有显著差异,在不同的光源下相同浓度溶液的光解动力学也有显著差异。在汞灯下,随pH增大,CA4P溶液光解速率变大;在日光灯下,随pH增大,CA4P溶液光解速率变小。CA4P水溶液的光解速率常数(k=0.0027h-1)小于CA4P甲醇溶液的光解速率常数(k=0.0058h-1)。β-CD、HP-β-CD、Tween80对CA4P溶液的光解速率常数影响:kblank(1×10-6L.mol-1.h-1)>kHP-β-CD(7×10-7L.mol-1.h-1)=kTween80(7×10-7L.mol-1.h-1)>kβ-CD(6×10-7L.mol-1.h-1)。结论光源、浓度、pH、溶剂对CA4P溶液的光解过程有显著影响,β-CD、HP-β-CD,Tween80对CA4P溶液的光解过程有一定抑制作用。 OBJECTIVE To study photolysis kinetics of eombretastatin A4 phosphate solution. METHODS CA4P solution was exposed to different light. Samples were determined by HPLC. RESULTS The photolysis kinetics of CA4P solution at different concentration under the same light was different. The photolysis kinetics of CA4P solution at the same concentration under different light was different. Under mercury lamp, photolysis rate was increased with pH increase;under daylight lamp, photolysis rate was decreased with pH increase. The photolysis rate constant of CA4P in water was smaller than that in methanol, respectively (k = 0. 002 7 h^-1 and k =0. 005 8 h-l ). The effect offl-CD,HP-fl-CD,Tween 80 on the photolysis rate constant of CA4P was different,the sequence was as follows:kblank ( 1×10^-6 L·mol^-1·h^-1) 〉 kHP-β-CD (7×10^-7L·mol^-1·h^-1 ) = kTween 80 (7×10^-7L·mol^-1·h^-1 ) 〉 kβ-Cd(6×10^-7L·mol^-1·h^-1 ). CONCLUSION Light source, concentration, pH and solvent show important influence on photolysis kinetics of CA4P. fl-CD,HP-fl-CD and Tween 80 to some degree inhibits photolysis process of CA4P.
