瘦素在实验性大鼠肝纤维化发生中的作用机制初探 被引量:5

摘要 目的通过检测肝纤维化大鼠血清及肝组织中瘦素(Leptin)的表达水平,初步探讨瘦素在大鼠肝纤维化发生中的作用机制。方法32只雄性SD大鼠,皮下注射四氯化碳(CCl4)油剂制备肝纤维化模型,分别于注射后1、4、8周处理动物,抽取血清并留取肝组织,采用放射免疫学方法检测血清中瘦素的表达水平,应用免疫组化方法检测肝组织中瘦素及转化生长因子β1(Transforming growth factorβ1,TGF-β1)的表达。结果放免及免疫组化结果均显示,与正常对照组相比,注射CCl4诱导后,肝纤维化大鼠血清及肝组织中瘦素的表达水平明显升高(P<0.01,<0.05),且1,4,8周组瘦素的表达强度呈明显递增趋势(P<0.01,<0.05)。瘦素主要表达于肝星状细胞胞质中。结论瘦素主要表达于肝星状细胞,并可能通过窦内皮细胞发挥重要的促纤维化作用。 Objective To explore the role of Leptin in the mechanism of liver fibrosis via investigating the expression of Leptin in rats with liver fibrosis. Methods Liver fibrosis models of rats were made by subcutaneously injecting with CCl4. Thirty two male SD rats ( weight 250 - 300 g) were randomly scarified for 1,4and 8 weeks after injection of CCl4 respectively, and their liver tissues were used for examining Leptin and TGF-β1 by immunohistochemistry stain. RIA was used to detect the level of serum Leptin in all groups rats too. Results The expressionof Leptin in rats after injection with CCl4 were found chiefly in hepatic stellate cells(HSC) and all significantly higher than those in normal rats ( P 〈 0.01, 〈 0.05 ). Moreover, it present with progressive tendency for the expression of Leptin in rats respectively at 1 st ,4th and 8th week after injection with CCl4 ( P 〈 0.01, 〈 0.05 ). Conclusions The expression of Leptin in HSC may play an important role in liver fibrogenesis of rat.
