摘要
目的建立超声波提取湖北地黄多糖的最佳工艺和测定湖北地黄各部分中多糖的含量。方法对超声波提取湖北地黄多糖的条件进行了研究并用蒽酮-硫酸法测定了湖北地黄各部分的多糖含量。结果由正交实验得出超声波提取的最佳条件为:湖北地黄与水的质量比为1∶40,提取温度为40℃,提取时间为30 m in。采用蒽酮-硫酸法于624 nm比色,得湖北地黄中各部分多糖含量为:根、茎、叶分别为15.74%,8.36%,4.21%。结论湖北地黄的多糖依据不同提取方法、不同部位含量有较大差异。
Objective To establish the optimum process of the polysacchafides extraction in seeds of Rehmanniaglutinosa Libosch, in Hubei province by ultrasonic wave technology and content determination of polysaccharides. Methods The extraction conditions with ultrasonic wave were studied and the contents of polysaceharides were determined by the anthrone - H2SO4 colorimetric method. Results The optimum process of the technology was abtained through orthogonal test:the ratio of[ S ]/[ L ] was 1: 40, the extraction temperature was at 40℃ and the extraction time was 30 min. The contents of the polysaceharides were determined by anthrone- H2SO4 colorimetric method (λ = 624 nm), and the contents from root, stem and leaf were 15.74%, 8.36%, 4.21% respectively. Conelusion The content of the polysaccharides in Rehmannia glutinosa Libosch. of Hubei province were different with different extraction mothed and in different part.
