摘要
目的:研究缬沙坦对糖尿病大鼠肾小管上皮细胞转化生长因子β1与结缔组织生长因子表达的影响。方法:将60只W istar大鼠随机分为对照组(n=20)和糖尿病模型组(n=40),糖尿病模型组给予一次腹腔注射链脲佐菌素制造糖尿病模型,模型成功后随机分为糖尿病组(n=20)和缬沙坦组(n=20);缬沙坦组每日给予缬沙坦30 m g/kg的剂量灌胃;第8周测定各组大鼠尿蛋白,处死大鼠取肾组织石蜡包埋切片,免疫组织化学法观察肾小管上皮细胞转化生长因子β1与结缔组织生长因子表达并进行半定量分析。结果:糖尿病大鼠尿蛋白排泄显著高于对照组(P<0.01),肾间质损害明显,糖尿病组大鼠肾小管上皮细胞转化生长因子β1与结缔组织生长因子阳性表达相对面积分别为0.2209±0.0405和0.1835±0.0611,与对照组比较均显著上调(P<0.01);缬沙坦组大鼠尿蛋白排泄较糖尿病组显著下降,其肾小管上皮细胞转化生长因子β1与结缔组织生长因子阳性表达相对面积分别为0.1435±0.0322和0.1183±0.0376,较糖尿病组显著下调(P<0.01)。结论:缬沙坦能显著下调糖尿病大鼠肾小管上细胞转化生长因子β1与结缔组织生长因子表达。
Objective To observe effect of valsartan on the expression on transforming growth factor-β1 and connective tissue growth factor in renal tubular epithelial ceils of diabetic rat. Methods Sixty Wistar rats were randomly divided into normal control group (n 20) and diabetic mellitus models group (n=40). Diabetic mellitus models were established by intraperitoneal injection of 60 mg/(kg · d) streptozotocin. After these models were successfully established, they were randomly divided into two groups, diabetic group who received 2.0 mL water for lavage and valsartan group who received 30 mg/(kg · d) valsartan for lavage. After eight weeks, the excretion of urinary protein was measured. The transforming growth factor-β1 and connective tissue growth factor in renal tubular epithelial ceils were detected with immunohistochemistry of SP methods. Results The excretion of urinary protein in diabetes group was significantly higher than in normal group. The positive areas of transforming growth factor-β1 and connective tissue growth factor in renal tubular epithelial ceils of diabetes group were 0. 2209±0. 0405 and 0. 1835±0. 0611, which were significantly higher(P〈0.01) than normal control group. In valsartan group, the positive areas of transforming growth factor-β1 and connective tissue growth factor in renal tubular epithelial ceils were 0. 1435±0. 0322 and 0. 1183 ±0. 0376, which were significantly lower than diabetic group (P〈0.01). Conclusion Valsartan can down-regulate the expression of transforming growth factor-β1 and connective tissue growth factor in diabetic renal tubular epithelial ceils.
