出血性视网膜脱离后兔眼视网膜变化及凋亡相关基因bax在其中的表达 被引量:2

摘要 目的观察出血性视网膜脱离(hemorrhageretinaldetachment,HRD)对视网膜的影响和凋亡相关基因bax在其中的表达,探讨凋亡在出血性视网膜脱离中的作用。方法21只青紫蓝兔42眼,随机平均分为2组,实验组经玻璃体腔向视网膜下注入自体抗凝血0.2mL,对照组视网膜下注入含肝素钠生理盐水0.2mL。2只青紫蓝兔的4眼作为正常对照组。分别在手术后1h、1d、3d、7d、10d、14d、28d对视网膜进行常规HE染色切片,观察细胞丢失情况及测量视网膜神经感觉层厚度。采用免疫组化染色和原位杂交检测方法观察bax在视网膜中的表达情况。结果实验组10d、14d、28d的平均视网膜神经感觉层厚度、视网膜神经节细胞数明显小于对照组(P<0.05)。HRD后1~7d的bax表达显著,其中在3d达到峰值,实验组1d、3d、7d的bax阳性细胞较对照组明显增多(P<0.001)。结论HRD较单纯视网膜脱离的视网膜损害严重,且有时间依赖性和呈现递进的损伤过程,凋亡相关基因bax的表达说明凋亡可能是HRD视网膜损伤的重要机制。 Objective To observe the morphological changes and the ex- pression of bax in a rabbit model after hemorrhagic retinal detachment (HRD) ,and to investigate the role of apoptosis in HRD. Methods Fourty-two eyes of 21 rabbits were divided into two groups randomly. In experimental group, HRD models were established with subretinal injection of autologous blood 0.2 mL,and in the control group, the heparine sodium was injected correspondingly. Four eyes of other 2 rabbits were set as the normal control group. The number of retinal ganglion cells and the thickness of the inner retinal layers were measured by HE staining method 1 hour and 1 day, 3 days, 7 days, 10 days, 14 days and 28 days after the establishment of HRD and RD, respectively. The expressions of bax gene protein and mRNA in the models of HRD and RD were evaluated with immunohistochemistry and in sttu hybridization. Results Ten days, 14 days and 28 days after the establishment of models, the thickness of the inner retinal layers and the number of retinal ganglion cells in experimental group were obvious fewer than those in the control group(P〈0.05). There were positive cells one day after establishment of HRD. There were significant expressions of bax 1 day to 7 days after HRD and the positive cells peaked on the 3rd day. On the 1st day, 3rd day, 7th day, the number of positive cells in experimental group were more than that in the control group(P 〈0.001). Conclusion The retinal damage of HRD were sever- er than that of RD, and showed a time-dependent manner and forward process. The expression of bax implicited that apoptosis might play an important role in mechanism of retinal injury after HRD. [ Rec Adv Ophthalmol 2006; 26 (7) : 495-498 ]
