Detection of Serum Protein Biomarkers by Surface-Enhanced Laser Desorption and Ionization Time of Flight Mass Spectrometry(SELDI-TOF-MS) in Patients with Lung Adenocarcinoma

摘要 [目的]用表面增强激光解析离子化飞行时间质谱(SELDI-TOF-MS)技术寻找肺腺癌病人血清蛋白标志物,探讨吸烟与肺腺癌血清中蛋白质组谱变化的关系。[方法]收集肺腺癌病人和正常对照人群血清各50例,按照年龄、性别、吸烟史进行一一配对,采用WCX2芯片技术对血清蛋白进行捕获,用蛋白芯片阅读器PBSⅡ对芯片进行扫描、分析。[结果]肺腺癌病人血清蛋白图谱与正常对照组相比,存在10个差异表达蛋白(P<10-5),其中包括6个高表达蛋白,分子量分别是4050Da、4205Da、5329Da、5899Da、7761Da、9285Da,4个低表达蛋白,分子量分别是2564Da、2932Da、3082Da、3442Da。肺腺癌病人有吸烟史的与不吸烟的进行比较,血清蛋白表达差异无显著性(P>0.01)。[结论]SELDI-TOF-MS技术能筛选出灵敏度和特异性较好的差异蛋白,为肺腺癌的早期发现提供了一种新的、无创实验方法。 [Purpose]To search specifie protein biomarker by SELDI-TOF-MS in lung adenocarcinoma patients and to investigate the relationship between smoking and protein biomarkers in lung adenocareinoma.[Methods] Fifty cases with lung adenocarcinoma and 50 controls were matched by gender ,age and bistory of smoking.WCX2 PoteinChip was used to capture serum protein followed by PBS Ⅱ analysis .[Results] Compared with normal conrtol,ten protein bomarkers were identified including six highly expressed potential protein biomarkers with relative molecular weights of 4050,4205,5329,5899,7761,9285Da respectively .Four low expressed potential protein biomarkers with relative milecular weights of 2564,2932,3082,3442Da respectively.There was no statistical difference in serum protein between smoker and non-smoker in patients with lung adenocarcinoma (P〉0.01).[Comclusion]Some potential protein biomarkers with higher sensitivity and specificty can be screened by SELDI-TOF-MS technique which provides a new and noninvasive method for early detecion of lung adenocareinoma.
