重组基因工程红细胞生成素对大鼠长期毒性实验的骨髓及血细胞观察 被引量:3

摘要 主要观察了重组基因工程人红细胞生成素(r-HuEPO)腹腔注射对大鼠长期毒性实验中血细胞和骨髓的刺激作用.实验分四组即溶剂对照组,生理盐水对照组,重组基因工程人红细胞生成素小剂量和大剂量组.连续给药3个月(每月测抗体1次)后,分别检测各组动物的血红蛋白、红细胞压积、血小板和骨髓等ll项指标.结果发现大.小剂量组的血红蛋白和红细胞压积均非常显著增高,骨髓中红系增生明显活跃,以早、中、晚幼有核红细胞为主,与对照组相比有显著差异(P<0.01 ),双核及核分裂多见.血小板增高,粒系增生相对受抑.停药30天后除部分骨髓象没有恢复正常外,血象指标均转为正常. In this study the stimulant effect on marrow of subcutaneous recombinant human erythropoi-etin(r-HuEPO) was investigated. Animals were divided into four groups: solvent control, saline control, low dosage and high dosage. After continuous administration for 3 months (antibody was detected once every month), blood pictures (including 11 items, such as Hb,Hct and Plat) and marrow of every group were observed respectively. The results showed that the level of Hb and Hct in the high dosage and the low dosage group was significantly increased and the prolifera-tion of red cell family was markedly improved (compared to control group, P<0. 01), especially for early erythroblasts. Binuclear and karyokinesis were seen commonly. The level of platelets was increased and the proliferation of granulocyte family was relatively inhibited.
