摘要 目的:通过实验建立能快速评价溶栓药疗效并与临床接近的实验动物模型。方法:应用导管从大鼠颈外动脉逆行至颈总动脉,注入已形成血栓。于手术后3、6、10、18h观察行为障碍并评分,分别取脑观察梗死灶及脑组织病理改变。结果:该模型行为障碍和脑梗死体积较为恒定,光镜下6h脑组织神经细胞开始肿胀,10h、18h病变严重。结论:该模型简便易行,对动物损伤小,重复性较好,可为脑缺血发病机制研究及临床脑血栓的治疗研究提供一种有效手段。 Objective To establish an experiment animal model of brain thrombus that can evaluate effect of thrombelysis drugs and adjacent to clinic. Methods Twenty-four Wistar rats were employed in this study. A cather was introduced into the external carotid artery (ECA) lumen into the lumen of internal carotid artery (ICA), and then injected the formed thrombosis. Behavior disorders were observed at 3, 6, 10, 18 h after the operation. Infarction and histopathologic changes were examined respectively. Results The model had stable behavior disorder score and infarct volume. Nerve cell swelled at 6 h after the operation, degeneration and necrosis of nerve ceil were severe. Conclusion The model has the advantages of easy operation, little damage to animal, preferable repetition character. It may be offer an available means for research about mechanism of brain ischemia and therapy of clinic brain thrombosis.
