谷氨酸对培养神经元的兴奋毒性及氯胺酮的保护作用 被引量:5

摘要 对16~18d胎龄的大鼠皮层细胞进行分离培养,观察过量(10μmol/L)谷氨酸对皮层神经元的影响以及氯胺酮的保护作用。结果表明:培养10d的神经元置于10μmol/L谷氨酸和低糖(lg./L)DMEM培养液中后,随着作用时间延长,乳酸脱氢酶(LDH)活性增加。如在谷氨酸处理前,于培养液中加入NMDA受体非竞争性拮抗剂氯胺酮,则LDH活性明显低于对照组。结果表明:10μmol/L谷氨酸能造成培养的神经元严重损伤,氯胺酮对此有明显的保护作用。 The toxic effect of glutamate on cultured cortical neurons and the protective effect of ketamine were studied on the culture of cortical neurons from 16 to 18-day-old fetal rats,The culture of 10 days was exposed to 10μmol/L glutamate and low glucose(1g/L).Dynamic observation at 10 min,3h and 24h showed that the release of lactate dehydrogenase(LDH)into the culture supernatant was a function of time,when the cell culture was pretreated with ketamine( 0~500 μmol/L)10 min beforehand,the neurotoxic effect of glutamate(10μmol/L at 3h)wasobviously decreased,These results demonstrate that the cultured corticalneurons from fetal rat are seriously damaged by 10μmol/L glutamate and such a damage is attenuated by ketamine,suggesting that ketamine may protect neurons from glutamate injury.
