摘要
目的研究白细胞介素1(interleukin1,IL-1)、肿瘤坏死因子α(tumornecrosisfactorα,TNF-α)对体外培养的小牛晶体上皮细胞(bovinelensepithelialcel,BLEC)增殖和胞浆内游离Ca2+浓度的影响,探讨它们在后囊混浊形成中的作用及其影响细胞增殖的机理。方法采用MTT比色法测定IL-1及TNF-α对细胞增殖的影响,用荧光指示剂Fura-2测定它们对胞浆内游离Ca2+的影响。结果IL-1102~105ng/ml、TNF-α102~104U/ml可显著促进细胞增殖,并增加胞浆内游离Ca2+浓度。结论推测IL-1、TNF-α参与了白内障术后的后囊混浊形成;它们影响细胞增殖的作用机理之一是通过胞浆内游离Ca2+。
Objective To study the effects of interleukin 1(IL-1)and tumor necrosis factor α(TNF-α)on the proliferation and cytosolic Ca^2+ of bovine tens epithelial cells(BLEC) in vitro. The effects of IL-1 and TNF-α on posterior capsular opacification (PCO) and the mechanism of effect on the proliferation were investigated. Methods The effects of IL-1 and TNF-α on the proliferation of BLEC were detected with MTT staining colorimetry, and cytosotic Ca^2+ was measured by fluoresence determination with Fura-2. Results IL-10^2- 10^5 ng/ml, TNF-α 10^2- 10^4 U/ml obviously could promote BLEC proliferation( P〈0. 05 - 0.01 ), and increase cytosolic Ca^2+ ( P 〈 0. 05- 0. 01 ) at theTNF-α on proliferation is by cytosolic Ca^2+.
