摘要
本文对已建的噬菌体抗体库中分离的人抗HBsAg克隆进行了序列分析和表达研究,发现19个克隆都具有相同的重链可变区基因,轻链可变区基因有4个相同,其余15个未进行序列测定.DNA序列分析表明 V_H、J_H分属V_HⅢ和J_H6,D对段为二个D基因的融合,V_k,J_k属于V_kⅠ和J_k4.构建了可溶性Fab段表达载体,发现在细菌培养上清和质周腔中有类似的表达,其表达量约为5ug/ml,与HBsAg可特异性结合,经测定其亲和系数约为0.5×10~9M^(-1).
Abstract Nineteen human anti-HBsAg clone isolated from a phage antibody library were analysed by DNA sequencing and restriction enzyme mapping. A single VH gene, and probably a single Vk gene were found. The VH and JH gene were derived from V H II and JH6. The D segment contained D-D fusion. Vh and Jk belonged to Vk I and JK4. Soluble Fab was expressed both in the bacterial culture supernatant and cell periplasm. The concentration of Fab in the supernatant was about 5ug/mnl. Fab produced by £ coli showed specific binding ability to HBsAg with the affinity of 0.5×109M-1
