用Nissl法复染Golgi法镀染的树鼠脑切片

摘要 Golgi染色法由于能充分显示神经元胞体及其突起而成为神经解剖学研究的重要手段之一,但此法镀染的神经元很少,不能定位。本实验用焦油紫对Golgi法染色的树脑切片进行复染。结果表明,Nissl法复染既能保留原来被镀染神经元及其树突的形态结构完整性,又可显示出原脑组织的细胞构筑特点及被镀染神经无所坚心的层次,克服了Golgi法不能定位的弱点。复杂的切片透明和反差效果好,不易褪色。 The Golgi′s staining method is one of the importanttechniques forneuroanatomy researches,because this method cansiqnificantly demonstrate the some and pro-cesses of a neuron. However, only a few neurons are impregnated by this method.Thus, SuchGolgi′sstaining method cannot show the locations of the neurons in the nervoussystem, In thisexpriment,Golgi impregnated sections of tree shrewbrains were cointerstained with cresyl vio-let .This Nisslcounterstaining not only can keep the integrity of the somas anddendrites of thepreviously impregnated neurons in the sections, butalso can demonstrate the cellular structuralfeatures of the originallyimpregnated brain tissues and the locations of the impregnatedneuronsin the tissues. Thus,Nissl counterstaining can overcome the drawbacksof Golgi′s stain-ing method which cannot dennot demonstrate thelocations of the neurons.The transparencyand constrast of thecounterstained sections are good and the sections do not fade easily.
