摘要
了解肿瘤坏死因子α(TNF-α)在人体疾病发生中的作用.方法:建立了TNF-α的双抗体放射免疫分析法(RIA).结果:方法的批内及批间CV分别为5.76%±1.73%和6.08%±1.91%.灵敏度为5.4pmol/L.临床应用结果显示,癌症、细菌感染及脑血管病病人的血清TNF-α值显著高于正常人(P<0.01),败血症、脑缺氧症新生儿的血清TNF-α值显著高于正常新生儿(P<0.01).结论:测定血清TNF-α值有助于临床疾病的诊断及治疗.
PURPOSE To gain insight into the role of TNF-α in human diseases. METHODS Radioimmunoassay of TNF-α was developed. Concentration of TNF-α was measured in the serum from healthy subjects and patients. RESULTS Intraand inter- assay CVs were 5.76% and 6.08% respectively. The detection limit of this assay was 5.4pmol/ L. There was a significant difference in the serum concentration of TNF-α between healthy subjects and patients with cancers,bacterial infections, cerebral vascular diseases, neonatal septicemia, neonatal hypoxia induced encephalopathy (P< 0.01). CONCLUSIONS Radioimmunoassay of TNF-α is useful for the study and diagnosis of many diseases.
