摘要
目的研究砂生槐子生物碱体外杀细粒棘球绦虫原头蚴的活性及其抗炎和对小鼠的急性毒性作用.方法用含生物碱(0.75～6.00 mg/ml)的1640培养细粒棘球绦虫原头蚴,0.03%亚甲基蓝拒染法检查原头蚴每天的死亡率;选择昆明小鼠,腹腔注射给药1次,生物碱浓度从150 mg/kg递增,连续观察14 d,记录死亡动物数,计算半数致死量;用二甲苯诱发小鼠耳肿胀动物模型,观察生物碱的抗炎作用.结果砂生槐子生物碱具有体外杀死原头蚴的药理活性,且杀虫作用随药物浓度的增加和作用时间的延长而增强.对小鼠的LD50=(207.81±20.82)mg/kg,对二甲苯诱发的小鼠耳肿胀无明显抑制作用.结论砂生槐子生物碱体外具有杀死原头蚴的活性,对小鼠具有中等强度的毒性,但对二甲苯诱发的小鼠耳急性炎症肿胀无抑制作用.
Objective To study the efficacy of killing Echinococcus granulosua protoscolex in vitro and anti-inflammatory effect, a-cute toxicity experiment in mice of alkaloids extracted from Sophora moorcroftiana seeds. Methods E. granulosua protoscolex was cultured with alkaloids at 0. 75-6. 00 mg/ml in RPMI-1640 and mortality was counted by methylene blue stain;Kunming mice were injected intraperitoneally with alkaloids to increase progressively from 150 mg/kg, mice had been observed for 14 days and LD50 result performed with NDST statistical software; Using the models of dimethylbenzene-induced mouse ear swelling, the anti-inflammatory effect of alkaloids was evaluated. Results The alkaloids extracted from S. moorcroftiana seeds (0. 75-6. 0 mg/ml) had obviously action of killing E. granulosua protoscolex in vitro. The insecticidal activity was gradually enhanced with a alkaloids variety of time-and-dose. LD50 for mice was (207. 81±20. 82) mg/kg. The alkaloids had no suppressive effect on mouse ear swelling by dimethylbenzene-induced. Conclusion The alkaloids has obviously insecticidal action in vitro and has midding intensive toxicity on mice, and no anti-inflammatory effect on mouse ear swelling by dimethylbenzene-induced.
