摘要
目的 观察吗啡依赖大鼠脑内海马区c fosmRNA水平的变化 ,研究糖皮质激素 (地塞米松 )控制阿片戒断症状的分子机理。方法 剂量递增腹腔注射吗啡建立大鼠成瘾模型 ,给与地塞米松干预 ,纳洛酮促瘾后观察戒断症状 ,应用原位杂交方法观察海马区c fosmRNA。结果 吗啡依赖大鼠经地塞米松干预后 ,由纳洛酮催促戒断症状评分明显低于未经地塞米松处理组 ;同时经地塞米松干预后 ,海马c fosmRNA的表达量明显低于未经地塞米松处理 ,但较盐水对照组高。结论 糖皮质激素(地塞米松 )能抑制吗啡依赖大鼠戒断症状以及海马c fosmRNA的表达。
Aim To explore between the molecular mechanism of glucocorticoid in the treatment of opioid withdrawal syndromes and the changes of c-fos mRNA expression.method Rats were administered with morphine by interaperitioneal injection with does adds to develop addiction mode.Twenty-four rats were randomly assigned into three groups and received different kinds of treatment (Group one:morphine,ip.Group two:morphine,dexamethasone,ip.Group three:saline,ip).Naloxone was administrated to precipitate withdrawal syndromes.In situ hybridization histochemistry (ISHH) was proceeded in hippocampus detect the changes of c-fos mRNA expression.Result The withdrawal syndromes in Group two were alleviated better than that of Group one,and was higher than that of Group three.Conclusion Glucocorticoid produces suppressing effect on the withdrawal syndromes of morphine dependent rats when the increasing expression of c-fos mRNA in hippocampus is inhibited.
